AhighlyreactiveserineproteasethatdisplaystheABIlitytodigestnativeproteins,therebyinactivatingenzymessuchasDNaseandRNasewithoutrecoursetoadenaturationprocess.Itisthemostpowerfulproteinaseamongallproteinasescharacterizedsofar.Itcleavesatthepeptidebondadjacenttothecarboxylicacidgroupofaliphatic,aromaticorhydrophobicaminoacids.
TheapplicationofthemoleculargradeProteinaseKissimilartotheNativeProteinaseKandisusedintheisolationorpreparationofhighmolecularweightnucleicacids.However,thisProteinaseKishighlypureandhasahigherspecificactivityandismorestableatroomtemperatureascomparedtonativeProteinaseK.
ItisstableandactiveoverawidepHrangeof4-12.
RecombinantProteinaseK isalsoactivewithSDS,ureaandEDTAandthemostactivetemperatureis65°C.
Itisinactivatedbydiisopropylfluorophosphates(DFP)andphenylmethanesulfonylfluoride(PMSF).
Application
• ThisRecombinantProteinaseKisextremelyeffectiveonnativeproteinsandcan,therefore,beusedtorapidlyinactivateendogenousnucleasessuchasRNasesandDNases.ThispropertymakesProteinaseKparticularlysuitablefortheisolationofnativeRNAandDNAfromtissuesorcelllines.
• Theenzymepromotescelllysisbyactivatingabacterialautolyticfactor.
• ProteinaseKisalsousedfortheanalysisofmembranestructuresbymodifyingproteinsandglycoproteinsoncellsurfaces.
• Theenzymeisparticularlywellsuitedforisolatingnucleicacidsforamplificationreactions.
• ProteinaseKcanbeusedtoremovecellulardebrisduringthepreparationofcolonylifts,andtotreattissuesectionstoensureefficientprobeinfiltrationduring insitu hybridization.
