LeupeptinisareversIBLecompetitiveinhibitorofserineandthiolproteases.Ithasbeenreportedtoinhibitcalpain,cathepsinB,cathepsinsHandLandtrypsin.Atypicalworkingconcentrationsisintherangeof10to100µM.Leupeptinappearstobeequallyeffectiveinanysaltform,adjustingforequivalentpeptidecontent.Thehemisulfatesaltswerethefirsttobecommerciallyavailable.Ofthethreesalts,thehydrochlorideistheleastinvasiveforminBIOLOGicalsettings.Noproblemorpreferenceforthetrifluoroacetate(TFA)formhasbeennotes;TFAisvolatile,socouldpossiblyberemovedbylyophilization.
Microbiallyproducedleupeptininhibitorwasfirstisolatedasamixtureoftwoverysimilarforms:acetyl-LeuLeu-Arg-alandpropionyl-Leu-Leu-Arg-al.Althoughthepropionylleupeptinisactiveasaninhibitortheacetylformismorecommonlyusedandisavailableinseveraldifferentforms.
Leupeptin,becauseofitsaldehydegroup,mayactasareducingagentandthereforeinterfereinproteindeterminationssuchasLowryand,toalesserextent,BradfordTheactivityofleupeptinsandrelatedanalogshasbeenstudied.
