Cycloheximideisahighlyeffectiveantibioticwithactivityagainstmold,yeast,andphytopathogenicfungi,withloweractivityagainstbacteria.Ithasbeenreportedtoinhibitthesynthesisofbothproteinsandmacromolecules,aswellasaffectapoptosisineukaryotes.InhibitionofproteinsynthesisisbelievedtobemediatedthroughDNAtranslationarrest,asdemonstratedinratthymocytes.Althoughcycloheximidehasbeenreportedtoinduceapoptosisinvariouscells,ithasalsobeenshowntoinhibitordelayinducedapoptosis.Theseobservationssuggestthatcycloheximide"seffectonapoptosismaynotbesolelythroughproteintranslationarrestandthatothermechanismsmaybeinvolved.CycloheximidehasalsobeenreportedtoinhibitFKBP12(peptidylprolylisomerasehFKBP12,PPIasehFKBP12)viacompetitiveinhibition.
