PHA-AandPHA-Bagglutinateerythrocytesindependentoftheirbloodgrouptype.Trypsintreatmentoferythrocytesdecreaseslectinbindingtothecellsurface.TheO-linkedglycopeptidereleasedbytrypsinisapotentinhibitorofboththeisolectins.Removaloftheterminalsialicacidresiduefromtheglycopeptideincreasesitsinhibitorypotency8-fold.Periodateorβ-galactosidasetreatmentofthetrypsinreleasedglycopeptidedestroysallinhibitoryactivity.Simplesugarsareverypoorinhibitorsoftheseisolectins.However,agalactoseresidueappearstoplayamajorroleintheselectinbindingmechanisms.ThesugarlinkageisalsoimportantsinceasyntheticN-linkedglycopeptideisnotinhibitory,whiletheO-linkedglycopeptidereleasedbytrypsinizationisapotentinhibitor.ThecellreceptorfortheisolectinshasthestructureGalactose-Gal-NAc-ser(orthreonine).
LectinthatrecognizesO-linkedgalactose-β-1,3-N-acetylgalactosamine,adisaccharide(Thomsen-FriedenreichantigenorT-disaccharide),presentoncellsurfaceglycoproteins.ABAcanalsobindgalactose-β-1,3-N-acetylglucosamine.ABAdoesnotbindmonosaccharides.ABAcanbeinternalizedbyclathrin-coatedvesiclesafterbindingtosurfaceglycoproteins.Afterinternalization,itinhibitsnuclearimportofnuclearlocalizationsignaldependentproteins.ABAinhibitsproliferationofmalignantcellswithoutcytotoxicityfornormalcells.ABAcanbeusedtorecognizeandpurifiyspecificglycoproteins,andforthepurificationofcellswithspecificsurfaceglycoproteins.ABAhaspotentialasanti-proliferativeagent.
