PNAdoesnotagglutinatenormalhumanerythrocytes,butstronglyagglutinatesneuraminidasetreatederythrocytes.LectinPNAisspecificforterminalβ-galactoseandbindspreferentiallytoacommonlyoccurringstructure,galactosyl(β-1,3)N-acetylgalactosamine.PNAhaspotentanti-Tactivitysimilartotheanti-Tantibodyinhumansera. PNAhasbeenusedintumourtissuedeterminationfortransitionalmucosamalignancies.Thelectinalsoagglutinatesneuraminidase-treatedhumanerythrocytesat<0.1µg/mlaftertrypsintreatmentofcellsanditsactivityisinhibitedbylactoseandgalactose. ThoughPNAdoesnotrequireanydivalentcationsforactivity,thepresenceofcalciumionsindiluentscanenhancethebindingofPNAtoreceptors,possIBLybyneutralizingthenegativechargesonsialicacidresiduesadjacenttothereceptorsequence.
PNAisusefulindistinguishingbetweennormalandtumortissuesandinassessingmalignancyintransitionalmucosa.Inaddition,PNAbindingcanbeusedtomeasurecellularmaturityinlymphoidtissues,todistinguishavarietyoflymphocytesubpopulationsinmanandexperimentalanimals,andtomeasurethelevelsoflymphoidcellpopulationsinmanydiseases.PNAcanbeemployedinthefractionationofstemcellsinmiceforuseinbonemarrowtransplantationacrosshistocompatibilitybarriers.
