ThisPCRGradeProteinaseKisparticularlysuitableforisolatingnucleicacidsforamplificationreactions,aswellasfortheanalysisofmembranestructuresbymodifyingproteinsandglycoproteinsoncellsurfaces.ItisextremelyeffectiveonnativeproteinsandcanthereforebeusedtorapidlyinactivateendogenousnucleasessuchasRNasesandDNasesduringnucleicacidisolation.ThispropertymakesProteinaseKparticularlysuitablefortheisolationofnativeRNAandDNAfromtissuesorcelllines.Itpromotescelllysisbyactivatingabacterialautolyticfactor.BecausetheenzymeistestedfortheabsenceofRNasesandDNases,andisvirtuallyfreeofDNA,itisespeciallysuitableforisolatingPCRandRT-PCRtemplates.ProteinaseKcanalsobeusedtoremovecellulardebrisduringthepreparationofcolonylifts,andtotreattissuesectionstoensureefficientprobeinfiltrationduringinsituhybridization.
