LeupeptinisareversIBLecompetitiveinhibitorofserineandthiolproteases.Ithasbeenreportedtoinhibitcalpain,cathepsinB,cathepsinsH,Landtrypsin.Typicalworkingconcentrationsareintherangeof10to100µM.Leupeptinappearstobeequallyeffectiveinanysaltform,adjustingforequivalentpeptidecontent.Thehemisulfatesaltswerethefirsttobecommerciallyavailable.Ofthethreesalts,thehydrochlorideistheleastinvasiveforminBIOLOGicalsettings.Noproblemorpreferenceforthetrifluoroacetate(TFA)formhasbeennoted;TFAisvolatile,socouldpossiblyberemovedbylyophilization.
Microbiallyproducedleupeptininhibitorwasfirstisolatedasamixtureoftwoverysimilarforms:acetyl-LeuLeu-Arg-alandpropionyl-Leu-Leu-Arg-al.Althoughthepropionylleupeptinisactiveasaninhibitortheacetylformismorecommonlyusedandisavailableinseveraldifferentforms. Leupeptin,becauseofitsaldehydegroup,mayactasareducingagentandthereforeinterfereinproteindeterminationssuchasLowryand,toalesserextent,Bradford.
